I find my surname sometimes written as Domagalski and sometimes Magalski. In my family tree research using DNA I am connected to Domagalski and Magalski. Are these two separate surnames ?

Does, “Do” proceeding a surname have any meaning or significance? As in some cultures an addition to a surname could mean, “son of” or some other meaning of identity.

I’ve research church records in South Bend Indiana and my great grandparents family has recorded births, baptisms etc..with Domagalski and sometimes Magalski.

Accordingly to "Hoffman's Polish surnames" Domagalski and Magalski are two separate surnames with different origins.

Surname Domagalski comes from Polish domagać się - "to demand"
